Methods Rats were intraperitoneally injected with TAA(10 mg/100 g rat weight dissolved in isosaline) or CCl4(100 μL/100 g rat weight diluted as 1:4 in corn oil) twice weekly for eight consecutive weeks to induce subchronic liver fibrosis. Blood and tissue samples were collected and analyzed. Results CCl4 but not TAA significantly decreased the RBCs, Hb, PCV, and MCV values with minimal alterations in other erythrocytic indices. Both hepatotoxins showed leukocytosis, granulocytosis, and thrombocytopenia. By the end of the experiment, the erythropoietin level increased in the CCl4 model. The serum iron, UIBC, TIBC, transferrin saturation%, and serum transferrin concentration values significantly decreased, whereas that of ferritin increased in the CCl4 model. TAA increased the iron parameters toward iron overload. RT-PCR analysis revealed increased expression of hepatic hepcidin and IL-6 m RNAs in the CCl4 model and suppressed hepcidin expression without significant effect on IL-6 in the TAA model. Conclusion These data suggest differences driven by hepcidin and IL-6 expression between CCl4 and TAA liver fibrosis models and are of clinical importance for diagnosis and therapeutics of liver diseases. 当归多糖对肝细胞Hepcidin表达的影响[J].中国老年学杂志,2018,第19期 目的探讨当归多糖(ASP)对人肝癌细胞Hep G2和人正常肝细胞L-02 Hepcidin表达的抑制作用。方法体外培养Hep G2和L-02细胞,RT-PCR检测不同浓度(0、100、200、400 mg/L)的ASP诱导不同时间(0、24、48、72 h和96 h)对两种细胞Hepcidin mRNA影响。Western印迹观察400 mg/L ASP诱导96 h对两种细胞Hepcidin表达的抑制作用。结果与对照组(0 mg/L)相比,浓度高于200 mg/L ASP能够显著下调Hep G2和L-02细胞Hepcidin mRNA表达(P<0. 001); 400 mg/L ASP连续作用48 h后对Hep G2和L-02细胞产生显著的Hepcidin抑制效应且抑制强度呈时间相关性; 400 mg/L ASP连续作用96 h后显著抑制Hep G2和L-02细胞Hepcidin表达(P<0. 001)。结论 ASP可以抑制Hep G2和L-02细胞中Hepcidin的表达。 Correlation of hepcidin content with iron metabolism, inflammatory response and immune response in patients with rheumatoid arthritis combined with anemia[J].Journal of Hainan Medical College,2018,第16期 Objective:To investigate the correlation of hepcidin content with iron metabolism, inflammatory response and immune response in patients with rheumatoid arthritis combined with anemia.Methods: A total of 50 patients with rheumatoid arthritis combined with anemia who were treated in our hospital between September 2014 and December 2017 were selected as as the study group and 48 patients with rheumatoid arthritis alone who were treated in our hospital during the same period were selected as the control group. The differences in serum levels of hepcidin (Hepc), iron metabolism indexes, inflammatory factors and immune response indexes were compared between the two groups of patients, and Pearson test was adopted to assess the correlation between Hepc content and disease activity.Results: Serum Hepc level of study group was higher than that of control group; serum iron metabolism index SF level was lower than that of control group whereas TF and TIBC levels were higher than those of control group; serum inflammatory factors IL-1α, TNF-α and MCP-1 levels were higher than those of control group whereas IL-4 and IL-10 levels were lower than those of control group; serum immune response indexes IgG and C3 levels were higher than those of control group. The serum Hepc content of patients with RA was directly correlated with the contents of iron metabolism indexes, inflammatory factors and immune response indexes.Conclusion:Hepc level abnormally increases in patients with rheumatoid arthritis combined with anemia, and it is directly correlated with the abnormality of iron metabolism, inflammatory response and immune response. Expression of fluorescent tagged recombinant erythroferrone protein[J].Asian Pacific Journal of Tropical Biomedicine,2018,第7期 Objective: To produce fluorescent tagged recombinant erythroferrone protein(ERFE_eGFP) for laboratory investigations. Methods: Erythroferrone(ERFE) gene was fused to green fluorescent protein(eGFP) gene and cloned in a pSecTag2Hygro plasmid. The constructed plasmid was amplified in Escherichia coli DH5α and the eGFP-fused ERFE(ERFE_eGFP) protein was expressed in human embryonic kidney(HEK293T) cell line. Results: The plasmid constructed from colony C6 contained ERFE_eGFP with the correct restriction sizes of 4.2 kb and expressed secretory ERFE_eGFP fusion protein(approximately size of 75 kDa) in HEK293T cell line. Conclusions: ERFE_eGFP recombinant protein is successfully expressed as a secretory functional protein and could be sensitively detected using fluorometry. This fusion protein might benefit future applications for localization of cellular ERFE receptors and competitive immunoassay of ERFE concentration.
